RNA-Protein interactions are important in regulating gene expression. Several RNA binding proteins associate with insulin mRNA and regulate its expression. Protein disulfide isomerase, PolyA binding protein and HuD interacts with 5ÚTR of insulin mRNA and regulate glucose stimulated insulin biosynthesis. These factors also associate PC1/3 and PC2 mRNA to regulate the expression of pPc1 and PC2 pro convertases that are required for processing of insulin to generate a functional insulin molecule. Altered expression of these regulatory molecules may lead to dysregulated insulin expression leading to insulin resistance. We also have identified Phosphatidyl inositol 5-phosphate 4-kinase (PIP4K) as an RNA binding protein. PIP4K was shown to be imported into malaria parasite, where it associates with specific transcripts. Multiple UUGU motifs present in the target RNA are essential for interaction with PIP4K. The RNA binding activity of the PIP4K is conserved across species and it associates with several different RNA.
